Since the emergence of COVID-19, the search for targeted therapies has been ongoing.
“Mechanistically, we had a conjecture in April and proposed a new idea that what cancer cells and virus-infected cells have in common is an overactive function of cell protein synthesis, which is how tumor cells and viruses survive natural selection.Many anticancer drugs do control overreplication by inhibiting transcription or translation.And homoharringtonine (HHT) had this mechanism, so we started three experiments in vitro and found that it could clear the virus, and we continued with the small animal experiments.Cell experiments demonstrated that HHT was present at extremely low concentrations (& LT;Novel Coronavirus can be completely cleared under 1 micromole).The virus clearance rate in the transgenic mice was up to five orders of magnitude.”Wu Zhongyi, a professor at the School of Life Sciences at Sun Yat-sen University, told First Finance.
In fact, wu Zhongyi and Wen Haijun, an associate professor at Sun Yat-sen University, proposed this scientific inference in April and published it in the “Outlook” column of the National Science Review.
HHT can inhibit the activity of DNA polymerase and the synthesis of nuclear protein, resulting in the disorder of DNA synthesis. It is a non-specific drug in the cell cycle, which has a strong killing effect on S-stage cells and a certain effect on G0 stage cells.It has an inhibitory effect on many kinds of tumors, and has no cross resistance with alkylating agents and purines.
Wu’s team found that HHT limits protein elongation and inhibits viral replication by interfering with the binding of tRNA to ribosomes. HHT is expected to reduce the viral load in patients very quickly, and its efficacy is relatively easy to assess.
“Coronavirus is unusual in that its genome encodes a ‘super protein’ (ORF1ab) longer than 8000 amino acids, which contains 16 predicted non-structural proteins (NSP) primarily responsible for novel Coronavirus replication.(we) speculate that a drug that blocks the protein extension will inhibit the replication by novel Coronavirus.The most effective of these drugs is HHT, which is widely used against leukemia and has been reported to inhibit other coronaviruses.”Wu zhongyi said.
Instead of launching clinical trials in China right away, Wu’s team followed the scientific path to in vitro and animal testing.
“In vitro and in mice, we found that the virus was completely cleared from the body three to five days after the drug was given.HHT showed a significant reduction in viral load of several orders of magnitude within a few days by aerosol administration.In vitro and in vivo data clearly justify the clinical trial plan.But such tests are only feasible in areas where there are many new infections.”Wu zhongyi said.
Based on the dose to treat leukemia, Wu’s team proposed a clinical trial of trichomonine, in which people weighing 60 kg were given a daily dose of 15 ml saline, atomized five times every four hours, and allowed eight hours of sleep.Each episode takes 15 minutes to atomize 3ml of solution.The total dose can be gradually increased to 4mg per day if safety permits.
“HHT has been used in humans for the treatment of leukaemia, and the safety, price and delivery methods are well established.”Wu zhongyi said.
